Katalin Kariko (Nobel 2023) and Drew Weissman's nucleoside modification breakthrough enabled COVID vaccines. Moderna ($100B+ peak valuation) and BioNTech (Ugur Sahin) are now applying mRNA to cancer (personalised neoantigen vaccines), flu, RSV, CMV, and rare diseases. The platform advantage: once you can make one mRNA drug, you can make any — the manufacturing is identical. The challenge: moving beyond vaccines to therapeutic proteins and gene editing delivery.
State of the art (2026)
mRNA in 2026 is two stories pulling in opposite directions. On the science, oncology is delivering: Moderna and Merck reported sustained 5-year Phase 2b data for intismeran autogene (mRNA-4157) plus KEYTRUDA at ASCO in June 2026 (49% reduction in recurrence or death in resected melanoma), with the Phase 3 INTerpath-001 readout still expected this year; BioNTech-Genentech autogene cevumeran showed durable T-cell memory in pancreatic cancer at AACR 2026. On the politics, the US is retreating: HHS wound down roughly $500m of BARDA mRNA programmes in August 2025, and the FDA issued a refusal-to-file on Moderna mRNA-1010 before reversing to an August 2026 PDUFA date. The combination vaccine path (mRNA-1083) was withdrawn and is being resubmitted. Personalised oncology, not respiratory vaccines, now carries the platform thesis.
